According to the clinical data published by various drugs, Tirzepatide is the king of weight loss. Two recently published phase 3 clinical data show that Tirzepatide can reduce weight by 26.6% in 84 weeks.
According to the latest results of SURMOUNT-3 and SURMOUNT-4 trials published by Eli Lilly, the subjects treated with Tirzepatide had a total average weight loss of 26.6% after 12 weeks of intensive lifestyle intervention and continued use of Tirzepatide for 72 weeks. If the patients treated with Tirzepatide do not forcibly control the use of drugs on time, the average weight loss can be 21.1%.
The results of this trial show that if Tirzepatide can be taken on time every day, the weight loss can reach 26.6%. If there is a certain amount of forgetting to take the medicine, the weight loss can also reach 21.1%. According to all the currently published data, Tirzepatide is the strongest weight loss drug in history.
In addition to Tirzepatide, the already marketed semaglutide and several clinical stage drugs also have good weight loss effects.
The world's first oral GLP-1 - semaglutide
According to clinical data released by Novo Nordisk in May this year, after 68 weeks of treatment, 50mg oral semaglutide (Rybelsus) can reduce weight by up to 17.4% (adherence to medication). If the patient stops taking the medication or forgets to take the medication for some reason, the weight loss effect can reach 15.1%.
Compared with injections, oral semaglutide saves some patients from the trouble of injections, which can improve patient compliance to a certain extent, and the weight loss effect is also good.
In addition to oral semaglutide, its injection Wegovy (2.4mg) can also be used for weight loss. According to clinical data released by Novo Nordisk in 2021, patients can successfully lose 16.9%-18.2% of their weight after 68 weeks of treatment.
Wegovy only needs to be injected once a week, and its weight loss effect is basically the same as that of the oral medication group.
Highest weight loss - Retatrutide
Retatrutide is a GLP-1/GIP/GCG triple receptor agonist developed by Eli Lilly, and it is also the world's first GLP-1/GIP/GCG triple receptor agonist drug to enter clinical phase 3.
Clinical phase 2 data showed that at 24 weeks, the average weight changes in the Retatrutide group were: 7.2% weight loss in the 1 mg group, 12.9% weight loss in the 4 mg group, 17.3% weight loss in the 8 mg group, and 17.5% weight loss in the 12 mg group.
At 48 weeks, the average weight changes in the Retatrutide group were: 8.7% weight loss in the 1 mg group, 17.1% weight loss in the 4 mg group, 22.8% weight loss in the 8 mg group, and 24.2% weight loss in the 12 mg group.
Retatrutide reduced the average weight of the subjects by 24.2% in just 48 weeks. It should be noted that Eli Lilly's telotriol also takes 84 weeks to reduce weight by 26.6%. According to industry media STAT, this is the drug that has achieved the highest weight loss so far.
If the drug is finally approved for marketing, it may break the weight loss record set by telotriol.

Masdutide is a GLP-1/GCG dual-target agonist jointly developed by Innovent Biologics and Eli Lilly. According to the clinical phase 2 data released by Innovent Biologics this year, after 24 weeks of treatment, the weight of the 9 mg group of Masdutide can be reduced by 15.4%, and the average change in weight from baseline is 14.7 kg lower than that of the placebo group. At the same time, the 9 mg group of Masdutide has a probability of 81.7%, 65.0%, 31.7% and 21.7% respectively, which makes the subjects' weight drop by at least 5%, 10%, 15% and 20% from baseline.
