Ruxolitinib is the first clinically available, potent, selective JAK1/2 inhibitor with an IC50 of 3.3nM/2.8nM in cell-free assays. The selectivity of JAK1 and JAK2 is more than 130 times higher than JAK3. Ruxolitinib kills tumor cells through toxic mitophagy. Ruxolitinib can induce autophagy and enhance apoptosis.
